補足情報

Rpt1 also known as PSMC2 or 19S proteasome regulatory subunit 2 is a protein involved in the ATPase activity within the 19S regulatory particle of the proteasome. This protein has a molecular weight of approximately 46 kDa and plays an important role in protein degradation. Rpt1 is expressed ubiquitously in various tissues reflecting its importance in cellular functions. The protein's structure includes an ATPase domain which is necessary for energy-dependent unfolding of substrates during proteolysis.
Biological function summary

This protein plays an important role in the function of the ubiquitin-proteasome system (UPS) a complex mechanism responsible for targeted protein degradation. Rpt1 is a component of the 26S proteasome specifically part of the 19S regulatory complex and it assists in the recognition and unfolding of ubiquitinated proteins before they are translocated to the 20S core particle for degradation. The protein's role in UPS is essential for maintaining cellular protein homeostasis by controlling protein levels and quality.

Pathways

The protein is integral to the ubiquitin-proteasome pathway which is essential for various cellular processes like cell cycle control signal transduction and stress response. Within this pathway Rpt1 works closely with other proteasome subunits and associated proteins such as ubiquitin and Rpn1. The protein's activity ensures the timely degradation of regulatory proteins and misfolded proteins allowing for regulation of protein quality and turnover within the cell.

Rpt1 has been implicated in neurodegenerative diseases such as Alzheimer's and certain types of cancers. The dysregulation of proteasome function involving Rpt1 may lead to the accumulation of protein aggregates associated with neuronal degeneration. In cancer alterations in proteasome activity mediated by Rpt1 and related proteins like Rpt2 can affect cell cycle progression and apoptosis evasion contributing to tumorigenesis. Understanding these connections provides insight into potential therapeutic targets within these diseases.
