    RECA-1 reacts with rat endothelial cell antigen and showed to be reactive with all vascular endothelium in three tested rat strains of different MHC-haplotype; Lewis (TR-11), BN (RT-1n) and OA (RT-1u). RECA-1 recognises a cell surface antigen. The antibody has been successfully applied in staining of viable endothelial cells in vitro, and of vascular endothelium in vivo. No reactivity of RECA-1 was seen with other cells e.g. fibroblasts, leukocytes and non endothelial stromal cells nor with other various tested species other than rat e.g. mouse, rabbit, sheep, goat and human. RECA-1 is a promising antibody for rat endothelial cell studies, and in particular for further defining nature and function of endothelial cell-specific antigens.

Inquiry: Dear abcam Your datasheet for ab22492 states that it has been updated on 6 september 2012. It still states that ab22492 is unsuitable for IHC-P and only for IHC-Fr. However, according to what I have read below, your answer to the customer's question is that ab22492 can be used for both IHC-P and IHC-Fr. Hence I am confused as to which I should believe - the updated datasheet for ab22492 or your reply to this customer's inquiry dated 31 May 2012. Pls clarify soon so I could buy the correct antibody from you.

Thank you for your query.

The epitope recognised by ab22492 is reported to be sensitive to formaldehyde fixation (which is commonly used in IHC-P). There are publications which have used this clone successfully in IHC-P but we have never been able to replicate their success in our labs. Because of this, we are currently unable to support the use of this product with paraffin sections.

I can suggest to use ab124258. This is the same antibody, but from a subclone of the producingmyeloma and this antibody is tested and guaranteed for IHC-P.

Inquiry: Dear abcam I am looking for an antibody that can be used in IHC-paraffin fixed Wistar rat liver tissue section to stain for endothelial cells, because I would like to observe the presence of blood vessels within the liver. Your product ab9774 seems to guarantee successful staining except for some queries that still are not addressed in the Q&A section. You cited Aleš Hejcl et al & Eugene Park et al using ab9774, and classified that journal as a success story for IHC-P. However, you have another antibody ab22492 that specifically stains anti-RECA1, and that journal could have used that antibody. Could you kindly clarify the matter above, and if ab9774 can indeed be used to stain rat liver endothelial cells? Thank you. Kind regards

ab9774 is a well characterized antibody is fully guaranteed to detect the endothelial cells. Please use the product without any hesitation.

ab22492 is also reactive against vascular endothelium so this product is also an option to try.

Thank you for your detailed reply.Lee et al must have used ab22492 for IHC-P, becausethey wrote under Fig 4 on page 576:

Figure 4 COMP-Ang-1 gene transfer attenuates the microvascular rarefaction and tissue ischaemia in the heart and kidney of SHRs. The microvasculature of... the kidney (C) was visualized and quantified by immunohistochemical staining with CD31 IgG on frozen sections or RECA IgG on paraffin sections, respectively.

The staining in their figure looks fantastic, so it must have worked really well for them. However, I havealready purchased ab9774 (as I needed a RECA-1 antibody urgently, and the specifications for ab9774 state that it works for IHC-P), and will be trying it out on my IHC-P sections next week.

I have just realised that ab9774 is not an anti-RECA1 antibody. RECA1 is the clone number of this anti-Endothelial Cell antibody.

The only 2 anti-RECA1 antibodies we have in our catalog are https://www.abcam.com/ab22942 and https://www.abcam.com/ab124258.

I am very sorry for the confusion. I would be pleased to offer you a replacement of ab9774 withone unit of ab22942 or ab124258. If you decide to try ab22942 in IHC-P we would be pleased if you could share the results with us so we could confirm if this antibody is suitable or not in IHC-P.
