RAB26 also known as Ras-related protein Rab-26 is a member of the Ras oncogene family. This protein has a mass of about 25 kilodaltons (kDa). Functionally RAB26 engages in intracellular membrane trafficking. Expression of RAB26 is prominent in the brain particularly in neuronal and glandular tissues where it plays an important role in vesicle transport and secretion processes.
Biological function summary

RAB26 facilitates the movement and regulation of secretory vesicles. It is part of the small GTPase complex a group of proteins involved in vesicle formation transport and fusion. RAB26 regulates the targeting and fusion of vesicles to specific sites within the cell contributing to the precise delivery of cargo like neurotransmitters and enzymes.

Pathways

RAB26 is critical in the exocytotic pathways that govern vesicular traffic and secretion. It closely interacts with proteins such as synaptotagmin and VAMP2 which are essential in synaptic vesicle fusion. Additionally RAB26 is involved in pathways that manage stress responses in cells interacting with proteins like RAB3 further modulating vesicle dynamics during cellular stress.

RAB26 has associations with neurological disorders including Parkinson's disease and some forms of epilepsy. These conditions often involve dysfunctional vesicular trafficking pathways. In Parkinson's disease RAB26 interacts with proteins like alpha-synuclein which accumulate and lead to neurodegeneration. In epilepsy irregularities in vesicle release processes implicate RAB26 and similar small GTPases. Understanding RAB26's role in these disorders helps to elucidate potential therapeutic targets.

The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different set of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion. Mediates transport of ADRA2A and ADRA2B from the Golgi to the cell membrane. Plays a role in the maturation of zymogenic granules and in pepsinogen secretion in the stomach. Plays a role in the secretion of amylase from acinar granules in the parotid gland.
