I am glad you found the reference useful, I am sorry it took a little while to get to you. Unfortunately, the experiment you are planning would not be covered by the Abpromise guarantee. This is because we have not ourselves tested the antibody in such an application (and have not recieved any information from our customers to support it) and would not therefore be able to guarantee how the antibody might perform.

The epitope mapping performed with the Anti-Prostate Specific Antigen antibody (ab403) can be found in the reference I have attached to this email.

The information can be found on page 7. The ab403 is of the clone number 8301 (ISOBM code #257) and has been mapped to the epitope group 3a. This is the region 158-163 of human Prostate Specific Antigen.

Please note that we currently have the clone number set to PSA1 but this will be updated. The clone number should be 8301.

As discussed on the telephone, I am sorry that as far as we are aware, ab403 PSA antibody has never been tested with samples from cow. All tested species covered by the guarantee are stated on the online datasheets, and these are updated as soon as any new information is brought to our attention.

I wanted to check the alignment of the immunogen with the bovine sequence. However, I am sorry to confirm that regrettably I have been unable to find the bovine sequence in the protein database sites. I would appreciate if it is possible for you to send the bovine amino acid sequence and I will be happy to do an alignment for you so we can predict whether or not this antibody may wok on cow samples.

I have found an antibody which may work in your experiments which I am able to share the immunogen information with you. The anti-Prostate Specific Antigen antibody [PSA1] antibody ab403 was raised against the native protein. However, epitope mapping has been performed to establish that the antibody binds to a region between residues 158-163 of the human protein.

You may therefore be able to synthesise this region of the protein to perform your experiment. Do you think this antibody might be suitable for your experiment? I would like to point out that we have not tried what you are hoping to use the antibody for and would therefore not be able to predict how it might perform.

I am waiting on a reference which details of how the epitope mapping was performed which may provide you with some useful information about different peptides used to probe the antibody with. Unfortunately, the person who has this information is not available until the middle of next week. Once I have this information I will pass it on to you.

All of the antibodies you have chosen are appropriate for the experiments you are hoping to perform. All of the antibodies have been raised against the human protein but the cross reactivity with the mouse protein has not been ascertained and I cannot therefore say whether they will also be able to detect the mouse form of the protein.

There are a few antibodies which may be worth considering as they may provide more experiments for the cost. All antibodies require optimisation before achieving the optimum results and it may therefore be that you will need to use more or less antibody than advised on the datasheets. The dilutions stated on the datasheets are guidelines only and will depend on a number of different factors.

For the anti-AMACR antibody you may want to consider ab63340. This is amouse monoclonal, provided at a volume of 100 ul at the same price as ab12498 but has been shown to be usable at a dilution of1/500 to 1/2000 compared to 1/25 to 1/50 for ab12498 (500 uL provided).

For the anti-Androgen Receptor antibody you may want to consider ab9474 (250 uL with a recommended dilution of 1/25-1/50). This should provide roughly the same number of experiments as ab47569 (100 uL with a recommended dilution of 1/50-1/100).

For the anti-Mitochondria antibody, you may want to consider ab3298. This is provided at a volume of 500 uL with a recommended dilution of 1/500-1/1000 compared to ab92824 for which 100 uL is provided at a recommended dilution of 1/1000.

For the anti-Prostate Specific Antigen antibody I was unable to identify another antibody which would provide you with more experiments.

In brief, according to the lab that performed this characterization: "Dilution series of antibody ab403 has been prepared and all dilutions have been incubated with 125 I (iodine)labeled PSA antigen. Bound antibodies have been separated with PEG precipitation and counts have been measured. Amount of free antibodies has been calculated by using total max of the test run. Affinity curve has been formed from values of bound/free versus bound and then the coefficient has been calculated using slope of the curve and molecular weight of the antigen." More detailed method descriptions can be found in the literature or on the internet. I recommend a book from the Practical Approach series: Immunoassays, edited by J. P.
