補足情報

Pma1 also known as plasma membrane H+-ATPase is an essential enzyme that functions as a proton pump in cells. It maintains a proton gradient across the plasma membrane by extruding H+ ions out of the cell which is an important process for cell homeostasis. Pma1 has a molecular mass of approximately 100 kDa. It is mainly expressed in the plasma membrane of yeast cells and other fungi where it plays a critical role in regulating intracellular pH and membrane potential.
Biological function summary

The plasma membrane H+-ATPase activity of Pma1 facilitates nutrient uptake and cell growth by providing an electrochemical gradient. In yeast it forms no known complex but works in conjunction with other membrane transporters to manage ions and metabolite exchange. Through this activity Pma1 indirectly supports various cellular processes including metabolism stress responses and signal transduction.

Pathways

The proton pump action of Pma1 is integral to the regulation of osmotic balance and energy metabolism pathways. It is linked to the glycolytic pathway by influencing the uptake of glucose through transporters. Pma1 is also associated with the oxidative phosphorylation pathway by maintaining the proton motive force necessary for ATP synthesis. It interacts with proteins such as glucose transporters and ion channels to ensure proper metabolic functions.

Pma1 is linked to the understanding of fungal infections as its function is critical for fungal cell survival. Inhibiting Pma1 can be a potential therapeutic target considering its absence in mammalian cells thereby providing specificity. It also offers insight into acidophilic disorders where proton gradients are disrupted. While no direct human protein counterparts exist its functional analogy in proton pumps relates it to the broader category of proton-pump-associated conditions.

ターゲットの情報

The plasma membrane ATPase of plants and fungi is a hydrogen ion pump. The proton gradient it generates drives the active transport of nutrients by H(+)-symport. The resulting external acidification and/or internal alkinization may mediate growth responses.
