  • 機能

    This is a calcium-independent, phospholipid-dependent, serine- and threonine-specific enzyme. Essential for T-cell receptor (TCR)-mediated T-cell activation, but is dispensable during TCR-dependent thymocyte development. Links the TCR signaling complex to the activation of NF-kappa-B in mature T lymphocytes. Required for interleukin-2 (IL2) production.
    PKC is activated by diacylglycerol which in turn phosphorylates a range of cellular proteins. PKC also serves as the receptor for phorbol esters, a class of tumor promoters.

    Inquiry: Dear Abcam, I'm writing to you as a PhD student at the University of Birmingham. I have recently purchased the Anti PKC theta (phospho T538) ab63365 antibody and would very much like to try it using the ELISA protocols. My main query is that I want to detect the amount of phospho T538 PKC theta from cells, so do you have any recommendations for a coating antigen I could use. I'm also unsure as to what I could use to prepare the standard curve. Any help you could offer would be greatly appreciated.

    In this case I would suggest using acapture ELISA assay. Here, the capture antibody will be an antibodyto capture total PKC theta and thedetection antibody will be yourab63365. For the capture antibody you can try https://www.abcam.com/PKC-theta-antibody-ab4133.html, although this has not yetbeen tested as capture antibody.

    Then you'll need a secondary antibody, conjugated to an enzyme for example, to detect this detection antibody. One example of a secondary antibody you could use is https://www.abcam.com/Rabbit-IgG-secondary-antibody-H-L-Pre-Adsorbed-ab7083.html
