Question

I was looking at an antibody at your website (cat.# ab32502) - anti-phospho-PKC-alpha (T638) rabbit antibody. I had a question about this product. Although the datasheet has a western blot image showing it's binding to PKC-alpha, is this antibody particularly tested for cross-reactivity with other members of PKC family like PKS-beta or PKC-delta.

Answer

We have not yet tested this item for cross reactivity against other PKC family members. BLASTs and immunogen alignment that I have performed show that this product, has 85% and 66% homology against mouse PKC beta and gamma respectively. Alignment against human show similar result, As such we would not expect reaction with PKC gamma however there is good change of reactivity against PKC beta. A review of the phospho sites T497, T638, S657, and Y658 of PKC alpha and gamma (our currently available phospho specific anti-PKC alpha products) show them to be fairly well conserved and it is likely that any product targeted to these sites of PKC alpha will recognize PKC beta as well.
