    Plays a central role during spermatogenesis by repressing transposable elements and prevent their mobilization, which is essential for the germline integrity. Plays an essential role in meiotic differentiation of spermatocytes, germ cell differentiation and in self-renewal of spermatogonial stem cells. Its presence in oocytes suggests that it may participate to similar functions during oogenesis in females. Acts via the piRNA metabolic process, which mediates the repression of transposable elements during meiosis by forming complexes composed of piRNAs and Piwi proteins and govern the methylation and subsequent repression of transposons. Directly binds piRNAs, a class of 24 to 30 nucleotide RNAs that are generated by a Dicer-independent mechanism and are primarily derived from transposons and other repeated sequence elements. Associates with primary piRNAs in the cytoplasm and is required for PIWIL4/MIWI2 nuclear localization and association with secondary piRNAs antisense. The piRNA process acts upstream of known mediators of DNA methylation. Participates to a piRNA amplification loop. Besides their function in transposable elements repression, piRNAs are probably involved in other processes during meiosis such as translation regulation. Indirectly modulate expression of genes such as PDGFRB, SLC2A1, ITGA6, GJA7, THY1, CD9 and STRA8. Inhibits tumor cell growth when repressed. When overexpressed, acts as an oncogene by inhibition of apoptosis and promotion of proliferation in tumors.

The protocol seems fine to me. However I have following questions;

- Did you try loading control? If yes, how was the results?
- Could you provide an image?
- In many cases no band problem can be overcome by incubating the antibodies over night or by decreasing the dilution. Could I suggest you to try the antibody overnight at 4C and 1/200 dilution if you haven't done so.

I hope these suggestions would be helpful. Please let me know if you have any question.

Inquiry: Good Morning, I have a problem with my Western Blotting: I used primary antibody Anti-piwil2 (ab 85084)but it seem does not identify positive control (mouse testis 30ug/ul). I incubate the membrane in blocking buffer (milk 5% in TBS with a minute percentage of detergent Tween 20),O.N, 4°C with gentle agitation on a shaker .After blocking wash the blot three times and added antibody Anti-piwil2 (dilute 1:500 with milk 5% in T-TBS O.N 4°C ). Could you give me suggestions to solve my problem?

I was wondering if the Abcam antibodies ab12337 and ab105393 (Piwil 1), ab85084 (Piwil 2) and ab87939 (Piwil 4) work on Platypus and Echidna. I would be doing Western blots and IHC with these antibodies. Also, if you have any information on any antibodies for Piwil 3 for the above applications and species, please pass them on. Please advise if we will be entitled to testing discounts as well.

All going well, we would eventually want to do CHIP-Seq with these antibodies. Hence, if you have any information on that too, can you please pass them on.

The sequence homology between human and Platypus species seems quite OK for antibodies ab12337 and ab85084 cross reactivity. These antibodies may be tried for further tests. We can provide abrreview discount for these products.
