  • 機能

    May affect the structure or silencing of chromatin by phosphorylating HP1 gamma/CBX3. Isoform 2 promotes the G1/S transition of the cell cycle via up-regulation of CDK2 activity and phosphorylation of CDKN1B, resulting in enhanced nuclear export and proteasome-dependent degradation of CDKN1B. Isoform 2 also represses CDKN1B transcription by phosphorylating and inactivating the transcription factor FOXO3. Plays a role in signal transduction in blood cells. Contributes to both cell proliferation and survival and thus provides a selective advantage in tumorigenesis.

      Immunoprecipitation - Anti-PIM1 antibody [EPR24447-39] (AB300453)
      PIM1 was immunoprecipitated from 0.35 mg A20 (mouse reticulum sarcoma B lymphocyte), whole cell lysate 10 ug with ab300453 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab300453 at ID63 dilution. VeriBlot for IP secondary antibody (HRP)(ab131366) was used at 1/5000 dilution.

      Lane 1: A20 (mouse reticulum sarcoma B lymphocyte), whole cell lysate 10 ug

      Lane 2: ab300453 IP in A20 whole cell lysate

      Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab300453 in A20 whole cell lysate

      Blocking and dilution buffer and concentration: 5% NFDM/TBST.

      Exposure time: 3 minutes
