Biological function summary
This toxin disrupts immune surveillance and modulates host immune responses to facilitate infection by Bordetella pertussis. Pertussis toxin is not an isolated agent but functions in concert with other virulence factors produced by the bacterium. This synergy potentiates its ability to impair neutrophil recruitment and function. Moreover PT impacts the ciliary activity in the respiratory tract contributing to the characteristic coughing in whooping cough by limiting the clearance of respiratory secretions.
Pathways
ADP-ribosylation by pertussis toxin directly affects the inhibitory G-proteins specifically Giα which play important roles in several signaling pathways including the cAMP signaling pathway. The toxin's ability to increase the levels of cAMP disrupts normal intracellular signaling leading to altered leukocyte behavior. Such disruptions extend its influence into pathways involving proteins such as adenylyl cyclase and phospholipase C which further modulate immune responses.
