Pertussis toxin also known as PT or PTx is a protein with an approximate mass of 105-115 kDa. The bacterium Bordetella pertussis expresses this toxin which consists of five subunits arranged in an A-B toxin structure. The A portion functions as the enzymatic domain while the B units facilitate binding to host cell receptors. Pertussis toxin executes its mechanical function by catalyzing the ADP-ribosylation of the Gi protein which inhibits signal transduction pathways by immobilizing the G-protein causing disruption in normal cellular communication.
Biological function summary

This toxin disrupts immune surveillance and modulates host immune responses to facilitate infection by Bordetella pertussis. Pertussis toxin is not an isolated agent but functions in concert with other virulence factors produced by the bacterium. This synergy potentiates its ability to impair neutrophil recruitment and function. Moreover PT impacts the ciliary activity in the respiratory tract contributing to the characteristic coughing in whooping cough by limiting the clearance of respiratory secretions.

Pathways

ADP-ribosylation by pertussis toxin directly affects the inhibitory G-proteins specifically Giα which play important roles in several signaling pathways including the cAMP signaling pathway. S1 is an NAD-dependent ADP-ribosyltransferase, which plays a crucial role in the pathogenesis of B.pertussis causing disruption of normal host cellular regulation. It catalyzes the ADP-ribosylation of a cysteine in the alpha subunit of host heterotrimeric G proteins. In the absence of G proteins it also catalyzes the cleavage of NAD(+) into ADP-ribose and nicotinamide. It irreversibly uncouples the G-alpha GTP-binding proteins from their membrane receptors.
