補足情報

This supplementary information is collated from multiple sources and compiled automatically.

The PACAP receptor also known as ADCYAP1R1 is a G-protein-coupled receptor that interacts with the neuropeptide PACAP (Pituitary Adenylate Cyclase-Activating Polypeptide). This receptor plays a role in transmitting extracellular signals into cellular responses. It is comprised of approximately 525 amino acids with a considerable molecular mass of around 59 kDa. The PACAP receptors are widely expressed in the central nervous system including regions like the hypothalamus hippocampus and cerebellum. It also appears in peripheral tissues such as the adrenal glands and testis.
Biological function summary

PACAP receptor plays significant roles in both neuronal and endocrine systems. It modulates neurotransmitter release and affects neurotrophic activities. The receptor often operates in a receptor-ligand complex with PACAP which enables it to participate in neuroprotective and neurite outgrowth functions. This receptor actively regulates activities within the hypothalamic-pituitary-adrenal axis impacting stress responses and circadian rhythms.

Pathways

Many processes involve the PACAP receptor through essential signaling pathways like the cAMP-PKA and MAPK pathways. These pathways integrate signals from multiple receptor types including glucocorticoid receptors. The PACAP receptor often interacts with other adenylyl cyclase-activating proteins enhancing its involvement in neural cAMP synthesis and related functions. This places the receptor within key pathways regulating brain development and neuroplasticity.

The PACAP receptor holds connections to both mental health and neurological conditions. Its abnormal function has associations with disorders such as anxiety-related disorders and migraine. The receptor's relationship with proteins like the CRH (Corticotropin-Releasing Hormone) receptor further implicates its role in the pathophysiology of stress-induced disorders. Targeting PACAP receptors can thereby offer therapeutic potential in the management of these conditions.

G protein-coupled receptor activated by the neuropeptide pituitary adenylate cyclase-activating polypeptide (ADCYAP1/PACAP) (PubMed : 32047270, PubMed : 33715378, PubMed : 35477937, PubMed : 36385145). Binds both PACAP27 and PACAP38 bioactive peptides (PubMed : 32047270, PubMed : 35477937, PubMed : 36385145). Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of downstream effectors. Activates cAMP-dependent pathway (PubMed : 32047270, PubMed : 33715378, PubMed : 35477937, PubMed : 36385145). May regulate the release of adrenocorticotropin, luteinizing hormone, growth hormone, prolactin, epinephrine, and catecholamine. May play a role in spermatogenesis and sperm motility. Causes smooth muscle relaxation and secretion in the gastrointestinal tract (PubMed : 32047270, PubMed : 33715378).
