    IHC: This antibody may be diluted to a titer of 1:25-1:50 in an ABC method.
    Staining Protocol: We suggest an incubation period of 30-60 minutes at room temperature.

      Is NSE sensitive to triton-X? Should I be leaving this out of my IHC protocol, or can I leave it in?

      Answer

      We recommend the use of 0.025% Triton in the TBS as it helps to reduce surface tension, allowing reagents to cover the whole tissue section with ease. It is also believed to dissolve Fc receptors, therefore reducing non-specific binding. Furthermore, TBS gives a cleaner background than PBS. Care must be taken using Triton when working with membrane-bound proteins as the Triton acts as a surfactant and strips the lipid component out of the membrane. Neuron specific enolase (NSE) is located in the cytoplasm of mature neurons so you should have no problem using Triton in your buffer.
