NMNAT3 or nicotinamide nucleotide adenylyltransferase 3 catalyzes the biosynthesis of NAD+ from NMN and ATP playing a role in cellular energy metabolism. Alternate names include NMNAT3p and Enzyme NIC. It has a molecular mass of around 31 kDa. NMNAT3 is expressed in a variety of tissues with significant expression in red blood cells and the mitochondria of other cell types.
Biological function summary

NMNAT3 participates in the NAD+ salvage pathway transforming NMN into NAD+ a fundamental coenzyme in cellular metabolism and redox reactions. It does not form part of any larger complex but functions independently within the cell. Its activity supports energy production DNA repair and cell signaling processes making it an important player in maintaining cellular homeostasis.

Pathways

The salvage pathway for NAD+ biosynthesis relies heavily on NMNAT3's enzymatic action. It works alongside related proteins like NMNAT1 and NMNAT2 which catalyze similar reactions in different cellular locations. This pathway intersects with the oxidative phosphorylation process a critical energy-producing pathway within mitochondria.

NMNAT3 has implications in conditions such as anemia and mitochondrial dysfunction. Altered expression or mutation of NMNAT3 may contribute to anemia by affecting red blood cell metabolism and NAD+ homeostasis. The protein also relates to neurodegenerative disorders due to its role in mitochondrial function and energy balance aligning with proteins like Sirtuins which also regulate NAD+ levels and have implications in cellular aging processes.

ターゲットの情報

Catalyzes the formation of NAD(+) from nicotinamide mononucleotide (NMN) and ATP (PubMed : 16118205, PubMed : 17402747, PubMed : 26616331). Can also use the deamidated form; nicotinic acid mononucleotide (NaMN) as substrate with the same efficiency. Can use triazofurin monophosphate (TrMP) as substrate. Can also use GTP and ITP as nucleotide donors. Also catalyzes the reverse reaction, i.e. the pyrophosphorolytic cleavage of NAD(+). For the pyrophosphorolytic activity, can use NAD(+), NADH, NaAD, nicotinic acid adenine dinucleotide phosphate (NHD), nicotinamide guanine dinucleotide (NGD) as substrates. Fails to cleave phosphorylated dinucleotides NADP(+), NADPH and NaADP(+). Protects against axonal degeneration following injury (PubMed : 16118205, PubMed : 17402747). May be involved in the maintenance of axonal integrity (By similarity). Also functions as a stress-response chaperone protein that prevents toxic aggregation of proteins; this function may be independent of its NAD(+) synthesis activity (PubMed : 18344983).
