The NFATC4 protein has24 isoforms with a wide range of molecular weights. The predicted molecular weights are approximately8, 20, 36, 47, and there are alsomany isoforms between 77-100 kDa. The full list of known isoforms can be found at this link:

The expression of the different isoforms may vary between species and tissues. This antibody has been used in WB in PubMed ID: 19828810, so that reference may also be able to provide some information about what size bands were observed. Based on the immunogen, we would expect this antibody to recognize isoforms 1 through 21. It is likely that thebands you are seeing between 60-80 kDa arespecific.

The western blot conditions will be standard: denatured and reduced samples, block the membrane with either 5% milk or BSA, incubate with antibody diluted 1/500-1/1000 overnight at 4C or one hour at room temperature. We know of one reference which used the antibody for western blotting: Abdul HM et al. Cognitive decline in Alzheimer's disease is associated with selective changes in calcineurin/NFAT signaling. The reactivity of the NFATC4 antibody ab62613 was tested by ELISA against the immunogen sequence, which was not phosphorylated at serine 676, and it was also tested for reactivity with the same sequence but phosphorylated at S676. Reactivity was the same in each case, so the antibody will react with total NFATC4, regardless of whether it is phosphorylated at S676.
