      Ly-6B.2 is a ~25-30 kDa GPI-anchored, heavily glycosylated protein expressed on neutrophils, inflammatory monocytes and some activated macrophages. High levels of expression are seen in bone marrow, spleen, lung and lymph nodes. N-glycanase treatment of thioglycollate elicited peritoneal neutrophil lysates lowers the apparent molecular weight of Ly-6B.2 to ~15 kDa.

      ab53457 has been successfully used for the immunomagnetic depletion of neutrophils during the enrichment of primitive hematopoietic cells from bone marrow and the depletion of myeloid cells in vivo.

      Thank you for contacting us this afternoon and reporting the problems encountered with the most recent lot of the Anti-Neutrophil antibody [7/4] (ab53457). I am sorry to hear this antibody is not providing satisfactory results.

      As discussed over the phone, it may be that you received a faulty vial. This is regrettable and I apologise for any inconvenience caused to you.I have arranged for a replacement vial of the antibody to be sent. This is on the order number 1154657. This should reach you tomorrow.

      I have also attached a questionnaire so that I can collect a few more details of the problems encountered and the difference in staining observed between the different lots. This will greatly aid in investigating the problem further.

      If you could also fill it out for the other antibody where you say you have been seeing some high background in the staining and I can have a look to see if there are any suggestions I can make to improve the staining.

      As discussed over the phone, I have also issued a replacement of the Anti-F4/80 antibody [SP115] (ab111101). As this was purchased in August of last year, we would not normally offer to replace the antibody as this is well outside of the Abpromise warranty period. However, I have been willing to do this for you as a goodwill gesture. As agreed, if you could please submit an Abreview for the ab111101, detailing the antigen retrieval used and any advice that may be useful for future customers that would be very useful. As mentioned, detailed information of the experiment performed are not needed if you do not wish to share it before publishing in a journal. If you need help in this please do let me know.

      Thank you for your inquiry. We have not tested MCA771GA on the Vm/Dk mouse strain. We ran a literature search, butwere unable to find any publications in which customers have tried this antibody on this mouse strain either.
