Neurokinin A also known as substance K is a neuropeptide belonging to the tachykinin family. This peptide consists of 10 amino acids and has a molecular mass of approximately 1.13 kDa. Neurokinin A is expressed mainly in the central nervous system and the peripheral nervous system where it operates as a neurotransmitter and neuromodulator. The peptide shows significant expression in the brain especially in areas like the hippocampus striatum and neocortex. It is also present in peripheral tissues like the gastrointestinal tract and blood vessels.
Biological function summary

Neurokinin A influences numerous physiological processes including smooth muscle contraction and glandular secretion. It is not part of a complex but interacts directly with its receptor the neurokinin 2 receptor (NK2R) which is a G protein-coupled receptor. By binding to NK2R neurokinin A triggers various intracellular signaling cascades that affect cell functions such as proliferation migration and contraction. These actions make it an important player in modulating responses related to pain perception stress and inflammation.

Pathways

Neurokinin A participates in the tachykinin pathway which is important for signal transduction and neurotransmission in the nervous system. Within this pathway neurokinin A is closely related to other tachykinins like substance P and neurokinin B sharing cross-talk through their respective receptors. This interaction underlies many physiological responses such as vasodilation bronchoconstriction and modulation of pain and stress responses. Neurokinin A also plays a role in the inflammatory response pathway influencing immune cell activation and cytokine production.

Neurokinin A has been linked to asthma and inflammatory bowel disease. In asthma neurokinin A contributes to bronchoconstriction and airway inflammation through its action on NK2R and interaction with substance P. In inflammatory bowel disease it can exacerbate inflammation by affecting the motility and secretion of the gastrointestinal tract where it often works in tandem with other inflammatory mediators like cytokines therefore impacting disease progression. Understanding its role in these diseases could lead to targeted therapies that modulate neurokinin A's activity.

Tachykinins are active peptides which excite neurons, evoke behavioral responses, are potent vasodilators and secretagogues, and contract (directly or indirectly) many smooth muscles.. Substance P. Is a ligand for TACR1, and triggers G protein-coupled receptor signaling via activation of phosphatidylinositol hydrolysis by phospholipase C. Substance P binding to TACR1 also triggers signaling via activation of adenylate cyclase activity which results in increased intracellular levels of cyclic AMP (cAMP) (By similarity). Is also a TACR3 agonist with low receptor affinity (PubMed : 37391393). Basic secretagogue neuropeptide released from the terminals of specific sensory nerves (PubMed : 30686732). Acts as a ligand for MRGPRX2 receptor in mast cells, initiating a signaling that mediates neurogenic inflammation and pain (PubMed : 30686732, PubMed : 34789875). Neurogenic inflammation includes mast cell activation, recruitment of immune cells and release of inflammatory mediators, such as cytokines and chemokines (By similarity). The inflammatory response can then activate or sensitize nociceptors, promoting pain (By similarity).. Neurokinin A. Is a ligand for TACR2, and triggers G protein-coupled receptor signaling via activation of G(q) and phosphatidylinositol hydrolysis by phospholipase C (PubMed : 35882833).
