NC2 alpha also known as Dr1-associated cofactor 1 (Drap1) is a transcriptional regulatory protein with a molecular mass of approximately 65 kDa. It functions mechanically as a repressor of RNA polymerase II transcription and is widely expressed in various tissues with higher levels noted in the liver and brain. NC2 alpha exerts its function by binding to TBP-DNA complexes modulating their transcriptional activities. Its role as a negative regulatory element is critical in controlling gene expression.
Biological function summary

NC2 alpha participates as part of the NC2 complex partnering with NC2 beta to fine-tune transcription initiation. The complex competitively interacts with TBP influencing the accessibility of transcription factors to the promoter regions. This regulation impacts a broad range of cellular processes which include cell cycle progression and differentiation. NC2 alpha through these biological functions plays a role in maintaining cellular homeostasis ensuring that genes are expressed at appropriate levels.

The association of the DR1/DRAP1 heterodimer with TBP results in a functional repression of both activated and basal transcription of class II genes. This interaction precludes the formation of a transcription-competent complex by inhibiting the association of TFIIA and/or TFIIB with TBP. Can bind to DNA on its own.
