      Immunocytochemistry/ Immunofluorescence - Anti-MZF1 antibody (ab64866)
      ICC/IF image of ab64866 stained MCF7 cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab64866, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

    Thank you very much for your reply.

    I have a supplementary question:

    I have found 3 different MZF1 isoforms on NCBI: NP_003413 (734aa), AAF80466 (485aa) and NP_001253962 (290aa).

    Aminoacid 3-15 in the 734 and 290aa is the same; AVLGSPDRAPPE. However, the 485aa (Not containing the SCAN domain) has aminoacids: PLVYAGFALQLG.

    Could you inform me the exact sequence of aminoacids the ab64866 is recognizing so that I know which of the isoforms I am seeing on the WB?

    I apologize as I have made an error in writing the amino acid number the correct one is from aa11 to 23; means the majority of the immunogen sequence is in RAPPEDEGPVMVK sequence of human MZF1. http://www.uniprot.org/uniprot/P28698

    Question

    Phone call: Customer kindly discussed the new lot being not working in chip. The current lot produced weak band with use of high concentration of antibody under same conditions. The previous lots worked very well in CHIP and western blot. Requested replacement of lot that works well in both applications.
