    • 機能

      Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity.

        Flow Cytometry - Anti-Myeloperoxidase antibody [2C7] (ab25989)
        Overlay histogram showing HeLa cells stained with ab25989 (red line). The cells were fixed with methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab25989, 1/20 dilution) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a decreased signal in HeLa cells fixed with 4% paraformaldehyde (10 min)/permeabilized in 0.1% PBS-Tween used under the same conditions.

      Question

      Inquiry: I am trying to create an MPO-DNA sandwich ELISA for immune complexes and I have been unsuccessful so far. I have been using a mouse polyclonal anti-human MPO antibody to coat, but I think this is the problem. I am interested in your monoclonal MPO Ab and think it might work but I can't afford to buy a full tube and have it not work for my experiment. I was wondering if there is any way you can send a small aliquot to test one of my samples? If it works then I would be interested in purchasing. I hope you understand and can help in some way.       For MPO, we have tested a pair of antibodies in sandwich ELISA and guarantee them to work in human samples. The 2 are listed below:

      Since sandwich ELISA's can be hit or miss with antibody pairing since each antibody needs to have a unique antigen that it recognizes, going with the pair above is what I recommend since we can guarantee they will work together while we do not guarantee that ab25989 will work with your other antibody. That way, if ab10165 and ab24467 do not work for you in sELISA and we cannot work with you to get them to work we will refund your purchase or send you replacements free of charge.

      I have an inquiry about your monoclonal myeloperoxidase (MPO) antibodies in general. We are going to be looking for possible MPO fusion proteins, most likely by looking for changes in MPO protein size by western blot. For this we would like a N-terminal MPO antibody. The only information that I could find about the epitope location for your MPO antibodies was that one is located in the heavy chain, which I believe starts at amino acid 279. What is the location of this epitope and do you have any antibodies that are more N-terminal than this one?

      Ab25989 recognizes an epitope on the heavy chain of myeloperoxidase. The protein is a multimer of 2 heavy chains (55kDa) and two light chains (15kDa), the heavy chains being linked by a disulfide bond. In Western blot under non-reducing conditions, the antibody recognizes native MPO (whole molecule) and will recognize the heavy chain following boiling of the sample.
