Reviewing this case, I would like to offer some suggestions to help optimise the results from ab74871.

1.) I agree with your customer that the band at 120kDa most probably is not in a complex since as your customer stated the samples are boiled and reduced. I can only speculate on the identity of this band, but would like your customer to do a no primary control to check if the secondary antibody is producing this band.

2.) I would like to suggest to decrease the boiling temperature to 65C and to prolong the heating time up to 10 min. Your target is a multi-pass membrane protein, and these proteins have the unfortunate tendency to aggregate during boiling periods with high temperatures and beta- mercaptoethanol due to their hydrophobic nature.

3.) Since the protein is expressed in the mitochondria, I can recommend to use a lysisbuffer that is a little harsher. When results are unpredictable, the detection is usually on its limits. Therefore it will work sometimes and sometimes not without apparent reasons. maybe a standard SDS lysisbuffer will help.

4.) Since rat is only a species that is predicted to react but not experimentally tested by us, I suggest to use a positive control that will deliver results like, for example, HeLa cell lysate.
