Anti-MICA + MICB 抗体 [6D4] (ab54413)

For GLIPR1, the immunogen for ab118382 is 97% homologous with XP_001117594 - the predicted protein sequence for Macaca mulatta. We would predict this antibody to work for monkey samples.

For GAB1 and MICA, we have antibodies that are predicted to react with non-human primate samples (ab104376and ab63709 respectively). This indicates that the sequence homology is greater than 95%.

For MICB, the mouse monoclonal antibody ab90237 was raised against the full length human protein, which shows 77% homology with Macaca mulatta (UniProt A4F5F7). Because this antibody is a monoclonal, it is difficult to predict whether the epitope will be located in a conserved region or not. Unfortunately, the other antibody we sell that recognize MICB will also cross-react with MICA: the 6D4 clone, available unconjugated (ab54413) and FITC conjugated(ab46529). This clone was also raised against the full length protein, and would therefore also be difficult to predict whether it will cross-react.

According to Aceview at NCBI these proteins are both be expressed in spleen (http://www.ncbi.nlm.nih.gov/IEB/Research/Acembly/av.cgi?db=human&term=mica&submit=Go&http://www.ncbi.nlm.nih.gov/IEB/Research/Acembly/av.cgi?db=human&c=Gene&l=MICB) . While we currently do not have any testing data this tissue type should be a quality positive control tissue.

Both MICA and MICB are widely expressed except within the central nervous system. When think abou positive controls however it may be best to use samples from areas of overexpression or from carcinomas. MICA and MICB are expressed in many, but not but not all, epithelial tumors of lung, breast, kidney, ovary, prostate and colon.MICA is overxpressed in the cortical and medullar epithelial cells of the thymus.

I would recommend using thymus slides as a positive control for IHC or cortical and medullar epithelial cells in ICC.If those are unavailable I would next recommend epithelial tumor cell from any of the tissues listed above.

We currently have two primary antibody / blocking peptide pairs to MICA for use in IHC-P:

Antibody ab62540 reacts with human and mouse, matching peptide: ab109170

Antibody ab93170 reacts with human, matching peptide: ab109171

Unfortunately, neither of these antibodies have been tested for cross-reactivity with MICB. I hope this helps, please let me know if you need any additional information.

Because this antibody was raised against C1R cells that were transfected with full length MICA, the epitope recognized by ab54413 may be located at any point on the protein. To our knowledge, the epitope for this antibody has not been mapped. For this reason, the only way to be certain to block the epitope would be to perform the competition assay using a full length MICA protein.

Using a full length protein in a competition assay is much less efficient than using a peptide and it can be very expensive to use enough protein to fully block the signal of interest. Unfortunately, in this case, using a short MICA peptide may not block the signal at all, depending on where the epitope is located.

Using MICA alone will fully block the signal of this antibody (even where it is binding to MICB) because it was immunized using only MICA.

While ab54413 will recognize both MICA and MICB, it was raised against MICA transfected C1R cells. For this reason, you could use a full length MICA protein (such as ab73819) as your positive control. We do not currently sell MICA + MICB as a single protein.I hope this helps, please let me know if you need any other information.
