Heat mediated antigen retrieval was performed using ab93684 (Tris/EDTA buffer, pH 9.0). ImmunoHistoProbe one step HRP Polymer (ready to use)was used as the secondary antibody. Negative control : PBS instead of the primary antibody. Hematoxylin was used as a counterstain.

ab14404 staining Mast Cell Tryptase in Mouse small intestine tissue sections by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Cells were fixed with paraformaldehyde, blocked with 5% serum for 60 minutes at 21°C and antigen retrieval was by heat mediation in 10mM Citrate buffer pH6.0. Samples were incubated with primary antibody (1/500 in PBS + 2% BSA) for 16 hours at 4°C. An Alexa Fluor^®488-conjugated Donkey anti-rabbit Ipolyclonal(1/400) was used as the secondary antibody.
