    ab54223 recognizes murine Ly-6C, a 131 amino acid ~14 kDa differentiation antigen, expressed on macrophage/dendritic cell precursors in mid-stage development (late CFU-M, monoblasts and immature monocytes), granulocytes, and on a wide range of endothelial cells and subpopulations of B- and T-lymphocytes.

    ab54223 is able to distinguish multiple mouse blood monocyte subsets: immature Ly-6Chi monocytes are recruited to acute peripheral inflammation and develop into Ly-6C+ exudate macrophages, whereas more mature Ly-6C-/lo monocytes are precursors for tissue macrophages and dendritic cells in steady state.

    ab54223 was originally described as recognizing a protein encoded by the LY6C gene. It has subsequently become apparent that the LY6C locus demonstrates polymorphism and the LY6C gene has been re-designated LY6C2. The LY6C1 gene encodes a similar protein with ~95% sequence homology to LY6C2

    Ly6C is a monocyte/macrophage and endothelial cell differentiation antigen regulated by interferon gamma, and may play a role in the development and maturation of lymphocytes. It is a member of the Ly6 multigene family of type V glycophosphatidylinositol anchored cell surface proteins. It is expressed on bone marrow cells, monocytes/macrophages, neutrophils, endothelial cells, and T cell subsets. Mice with the Ly6.2 allotype (e.g., AKR, C57BL, C57BR, C57L, DBA/2, PL, SJL, SWR, 129) have subsets of CD4+Ly6C+ and CD8+Ly6C+ cells, while Ly6.1 strains (e.g., A, BALB/c, CBA, C3H/He, DBA/1, NZB) have only CD8+Ly6C+ lymphocytes.

Tissue was fixed with paraformaldehyde and then blocked with TNB for 30 minutes at 22°C followed by incubation with the primary antibody, at a 1/100 dilution, for 18 hours at 4°C. A Biotin-conjugated goat anti-rat polyclonal was used as secondary antibody at a 1/500 dilution.

    Question

    How was this Ly6C monoclonal antibody developed? What is the target sequence?

    Is this antibody actually for CD59 or Ly6C? I thought these two molecules were different molecules, but the way the website describes is not clear, makes me wonder if this antibody is actually for CD59 https://www.abcam.com/ly6c-antibody-er-mp20-ab15627.html

    The reason I am asking is that a friend of mine from a different lab recently stained a tissue for Ly6C (clone ER-MP20), and got lots of endothelial staining, but very little immune cell staining if any. I just want to use Ly6C as a monocyte marker, but I realized that I did not even know the difference between CD59 and Ly6C. Could you clarify what this Ly6C antibody is for?

    The production and characterization of the ER-MP20 clone are discussed in the following reference.

    The authors' summary, at the end of the abstract, is that "the antigen detected by ER-MP20 is most likely identical to Ly-6C."

    The text on the datasheet of ab15267 (but not the ab54223 datasheet), in the General Notes section and the Primary antibody Notes sections, refers to CD59, giving the impression, as you point out, that the antibody is actually anti-CD59. This was a mistake on our part, and the text will be removed to prevent confusion in the future.

    We do not have an explanation for your colleague's observations of endothelial staining, but there is some evidence that Ly-6C is expressed on some endothelial cells, as discussed in the following reference.
