Immunofluorescence analysis of Lamin B1 using Anti-Lamin B1 antibody - Nuclear Envelope Marker. Lamin B1 was detected in an immunocytochemical section of U2OS cells. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent for 15 mins. The cells were blocked with 10% goat serum. And then incubated with 5 μg/mL ab65986 overnight at 4°C. DyLight®488 Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1 : 500 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 5 μg/mL ab65986 overnight at 4°C. Cy3 Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1 : 500 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

Immunoprecipitation of Lamin B1 in Hela whole cell lysate. Western blot analysis of Lamin B1 using Anti-Lamin B1 antibody - Nuclear Envelope Marker; Lane 1 : Hela whole cell lysates (30ug); Lane 2 : Rabbit control IgG instead of ab65986 in Hela whole cell lysate; Lane 3 : Anti-Lamin B1 antibody - Nuclear Envelope Marker (ab65986) (2μg) + Hela whole cell lysate (500μg). After electrophoresis, proteins were transferred to a membrane. Then the membrane was incubated with ab65986 at a dilution of 0.5 μg/mL and probed with a goat anti-rabbit IgG-HRP secondary antibody. The signal was developed using ECL Plus Western Blotting Substrate. A specific band was detected for Lamin B1 at approximately 70-72 kDa. The expected band size for Lamin B1 is at 66 kDa. The band at ~50kDa is IgG Heavy Chain.

Biological function summary
Lamin B1 serves many important functions by providing structural integrity and regulating gene expression through chromatin organization. Part of the nuclear lamina complex Lamin B1 interacts with other lamin proteins such as Lamin A and Lamin C forming a mesh-like structure that supports the nuclear envelope. This protein also affects cellular processes such as DNA replication RNA transcription and cell division indicating its broad impact on various cellular activities.
Pathways
Lamin B1 plays an important role in gene expression regulation and cell cycle control. It interacts with pathways involved in chromatin remodeling influencing the access of transcription factors to DNA which affects gene expression profiles. Lamin B1 associates with related proteins such as Lamin A/C and other nuclear matrix proteins to maintain cellular architecture stability and normal cell function. This organization is vital to ensure the accuracy of cellular processes and prevent anomalies during cell division.
