Histone methyltransferase that specifically trimethylates 'Lys-9' of histone H3. H3 'Lys-9' trimethylation represents a specific tag for epigenetic transcriptional repression by recruiting HP1 (CBX1, CBX3 and/or CBX5) proteins to methylated histones. Mainly functions in euchromatin regions, thereby playing a central role in the silencing of euchromatic genes. H3 'Lys-9' trimethylation is coordinated with DNA methylation. Probably forms a complex with MBD1 and ATF7IP that represses transcription and couples DNA methylation and histone 'Lys-9' trimethylation. Its activity is dependent on MBD1 and is heritably maintained through DNA replication by being recruited by CAF-1. SETDB1 is targeted to histone H3 by TRIM28/TIF1B, a factor recruited by KRAB zinc-finger proteins. -

This data was developed using ab300573, the same antibody clone in a different buffer formulation.
KMT1E / SETDB1 was immunoprecipitated from 0.35 mg K562 (human chronic myelogenous leukemia lymphoblast), whole cell lysate 10 ug with ab300573 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab300573 at 1000 dilution. VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution.
Lane 1: K562 (human chronic myelogenous leukemia lymphoblast), whole cell lysate 10 ug
Lane 2: ab300573 IP in K562 whole cell lysate
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab300573 in K562 whole cell lysate
Blocking and dilution buffer and concentration: 5% NFDM/TBST.
Exposure time: 128 seconds
Lysate was freshly made and used for Immunoprecipitation immediately to minimize protein degradation.The band beneath the target band (100 kDa) is likely to be degraded target fragments.
