    Catalytic subunit of the NuA4 histone acetyltransferase complex which is involved in transcriptional activation of select genes principally by acetylation of nucleosomal histones H4 and H2A. This modification may both alter nucleosome-DNA interactions and promote interaction of the modified histones with other proteins which positively regulate transcription. This complex may be required for the activation of transcriptional programs associated with oncogene and proto-oncogene mediated growth induction, tumor suppressor mediated growth arrest and replicative senescence, apoptosis, and DNA repair. NuA4 may also play a direct role in DNA repair when recruited to sites of DNA damage. Directly acetylates and activates ATM. In case of HIV-1 infection, interaction with the viral Tat protein leads to KAT5 polyubiquitination and targets it to degradation.

    Ubiquitinated by MDM2, leading to its proteasome-dependent degradation.

      KAT5 / Tip60 was immunoprecipitated from 10 μg HCT 116 (human colorectal carcinoma epithelial cell) nuclear fraction with ab300521 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab300521 at 1/1000 dilution. VeriBlot for IP secondary antibody (HRP) (ab131366), was used as secondary antibody at 1/5000 dilution.

      Lane 1: HCT 116 nuclear fraction 10 μg (Input).

      Lane 2: HCT 116 nuclear fraction.   

      Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab300521 in HCT 116 nuclear fraction.

      Blocking and dilution buffer: 5% NFDM/TBST.

      Exposure time: 3 minutes.

      Observed MW(KDa): 60, 50

      Flow Cytometry (Intracellular) analysis of HCT 116 (human colorectal carcinoma epithelial cells) labeling KAT5 / Tip60 with ab300521 at a 1/50 dilution (red). Cells were fixed with 4% paraformaldehyde and permeabilized with 90% methanol. A Goat Anti-Rabbit IgG (Alexa Fluor® 488, (ab150081)) was used as the secondary antibody at a 1/2000 dilution. Isotype control - Rabbit monoclonal IgG (ab172730) (black). Unlabelled control - Cells without incubation with primary antibody and secondary antibody (blue)
