KAT3 also known as Cbp/p300 is a lysine acetyltransferase enzyme which plays a significant role in chromatin remodeling and transcriptional regulation by acetylating histones and non-histone proteins. Its molecular mass is approximately 265 kDa. This enzyme is expressed in a wide range of tissues indicating its participation in various cellular functions. KAT3’s widespread expression supports its involvement in multiple physiological processes critical to cell survival and differentiation.
Biological function summary

KAT3 functions as a transcriptional coactivator and is part of several transcriptional complexes. It interacts with various transcription factors enhancing their ability to regulate gene expression. By acetylating target nuclear proteins KAT3 modulates chromatin structure thereby facilitating gene transcription. This protein manipulation enables cell proliferation differentiation and maintenance of homeostasis highlighting its diverse biological roles.

Pathways

KAT3 actively participates in the signaling pathways RAS/RAF/MEK/ERK and PI3K/AKT illustrating its influence in cell growth and survival. It frequently interacts with proteins such as CREB and AP-1 which are vital in activating transcription and response to environmental signals. KAT3’s integration into these pathways highlights its critical involvement in managing cellular processes responsive to extracellular cues.

KAT3 is closely linked to cancer and Rubinstein-Taybi syndrome. Mutations or deregulation in KAT3 expression or function often result in abnormal cell growth contributing to oncogenesis. In Rubinstein-Taybi syndrome genetic mutations affecting KAT3 hinder normal developmental processes. Throughout these conditions KAT3 interacts with p53 a pivotal protein in tumor suppression and cell cycle control accentuating its relevance in maintaining cellular integrity and preventing disease progression.

Catalyzes the irreversible transamination of the L-tryptophan metabolite L-kynurenine to form kynurenic acid (KA), an intermediate in the tryptophan catabolic pathway which is also a broad spectrum antagonist of the three ionotropic excitatory amino acid receptors among others. May catalyze the beta-elimination of S-conjugates and Se-conjugates of L-(seleno)cysteine, resulting in the cleavage of the C-S or C-Se bond. Has transaminase activity towards L-kynurenine, tryptophan, phenylalanine, serine, cysteine, methionine, histidine, glutamine and asparagine with glyoxylate as an amino group acceptor (in vitro). Has lower activity with 2-oxoglutarate as amino group acceptor (in vitro).
