      The cytoplasmic domain of the integrin-ß4 subunit recruits the adaptor protein Shc and is required for assembly of hemidesmosomes. Tyrosine phosphorylation of multiple sites within the cytoplasmic domain regulates these cellular events. In particular, tyrosine 1526 interacts with the phosphotyrosine binding domain of Shc and is required for Shc activation. In addition, tyrosine 1494 is required for integrin-mediated IRS-2 phosphorylation and activation of PI3-kinase. More importantly, this site is critical for integrin a6ß4 increases in carcinoma invasion.

      Integrin alpha-6/beta-4 is a receptor for laminin. It plays a critical structural role in the hemidesmosome of epithelial cells.

      Integrin alpha-6/beta-4 is predominantly expressed by epithelia. Isoform beta-4D is also expressed in colon and placenta. Isoform beta-4E is also expressed in epidermis, lung, duodenum, heart, spleen and stomach.

      Defects in ITGB4 are a cause of epidermolysis bullosa letalis with pyloric atresia (EB-PA) [MIM:226730]; also known as junctional epidermolysis bullosa with pyloric atresia (PA-JEB) or aplasia cutis congenita with gastrointestinal atresia. EB-PA is an autosomal recessive, frequently lethal, epidermolysis bullosa with variable involvement of skin, nails, mucosa, and with variable effects on the digestive system. It is characterized by mucocutaneous fragility, aplasia cutis congenita, and gastrointestinal atresia, which most commonly affects the pylorus. Pyloric atresia is a primary manifestation rather than a scarring process secondary to epidermolysis bullosa.
      Defects in ITGB4 are a cause of generalized atrophic benign epidermolysis bullosa (GABEB) [MIM:226650]. GABEB is a non-lethal, adult form of junctional epidermolysis bullosa characterized by life-long blistering of the skin, associated with hair and tooth abnormalities.

    thank you again for your help previously in designing the parameters to run a phospho-b4 integrin elisa. I tried to run this and had some problems, and I suspect it may be due to the lysis buffer I am using. Do you have any suggestions for what lysis buffer to use for this kind of a membrane protein? I am guessing that I want something that doesn't denature the protein, but releases it from the membrane fraction - would RIPA buffer work for this? Or do you have a suggestion of what other lysis buffer could work to release the phospho-b4 integrin and be compatible with an ELISA? Thanks!

    NP-40 and RIPA buffers are good for solubilizing membrane proteins. Please see our western blotting guide for some further details and buffer recipes at https://www.abcam.com/index.html?pageconfig=resource&rid=11375

    It might also be necessary to use the extraction buffer as a wash buffer. A typical wash buffer is just TBST or PBST. Washing with TBST or PBST may make the membrane protein "unhappy" such that it is not soluble anymore or does not retain expected function.

    Also be sure to include phoshatase inhibitors in the extraction buffer.

    I had a question regarding the products ab29044 and ab63546, both
    antibodies for phosphorylated b4 integrin, but phosphorylated at
    different sites. The tested application notes says that these have
    been tested for use in ELISAs, and this is exactly what I wanted to
    do. However, no further details are given - do you have the other
    products I would need to run such an elisa? i.e. prepared elisa plates
    or suggested capture antibodies and a protocol I could use to look for
    both of these? Please let me know, thanks!

    I have checked our catalog and unfortunately we don't have any beta 4 integrin ELISA kits available. Nonetheless I'm happy to help you choose antibodies and any other reagents you need that will help you set up your integrin beta 4 ELISA.

    1. What species of integrin beta 4 will you be working with?

    2. What sort of samples will you be using?

    3. Do your samples contain small amounts of integrin beta 4 such that sensitivity is really criticial or do your samples express large amount of integrin beta 4?

    4. Will you be comparing phosphorylated beta 4 integrin to total integrin?
