The larger precursor can be detected at approximately 26kDa. A smaller band can sometimes be observed at approximately 12.5kDa. This consists of multiple isoforms (this can appear smeared as they are slightly different molecular weights).

    Question

    Hello,

    We are using Abcam ab37158 (Rabbit polyclonal ab against human IL32) for IHC studies.

    We would like to also use it for western blot analysis with cell lysates.

    There are several options for lysate preparations with or without detergents.

    We would prefer a buffer without a detergent for a potentially wider range of applications.

    I would therefore like to know if the antibody prep will work with native protein preparations?

    Specifically, I will appreciate if you could share the conditions/protocol used for the generation of information shown in the antibody data sheet so that we could use it in parallel as a positive control.

    Thank you for your enquiry.

    I can confirm that ab37158 was tested under both reducing and denaturing conditions. Here is the standard protocol for sample sample preparation.

    Spleen tissue lysate was prepared by homogenization in modified RIPA buffer (150 mM sodium chloride, 50 mM Tris-HCl, pH 7.4, 1 mM ethylenediaminetetraacetic acid, 1 mM phenylmethylsulfonyl fluoride, 1% Triton X-100, 1% sodium deoxycholic acid, 0.1% sodium dodecylsulfate, 5 μg/ml of aprotinin, 5 μg/ml of leupeptin. Tissue and cell debris was removed by centrifugation. Protein concentration was determined with Bio-Rad protein assay. The product was boiled for 5 min in 1 x SDS sample buffer (50 mM Tris-HCl pH 6.8, 12.5% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue) containing 5% β-mercaptoethanol.

    We have not tried the antibody under native WB but as it works well in IHC and IF, there is a good chance it could detect the native protein in other assays.

    As you know we have ordered a number of anti-human ab from Abcam. The list
    is attached...
    I would like to request fro your assistance in the following...
    We would like to get the information about the recommended diluent for the
    ab to be used for IHC with paraffin embedded tissue sections since its known
    to affect results at times.
    Other information regarding antigen retrieval conditions etc will be
    available on the net, but could be refurbished if they are updated.
    Equally important is the source of positive and negative controls.
    I would especially like to request for slides of both the types of controls
    as they are vital to decide/judge the variations in the protocols. I hope
    this is done as it will serve us for a long time in future.
