        IHC image of IL-17A staining in mouse lymph node formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab91649, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

      Inquiry: I used this antibody on sections of mice skin (IHC-P) but I obtained staining only on blood vessels (I tried on lymph nodes and the staining was good). I can not quantify the secretion of IL-17 because of the nonspecific staining. Blocking: protein block serum free (dako) for 10 min Primary antibody: 1/400 and 1/1000 for 15min or 1h Antigen retrieval: citrate buffer (pH6) 23 min micro-waves Secondary antibody : Envision anit-rabbit for 30min at RT Wash steps with Tris (pH 7.6) Detection with DAB

      Thank you for your email. I am sorry to hear that you have been experiencing problems with this antibody.

      I have read the details you have kindly provided and have following further questions for better understanding of the problem;

      - We generally recommend antibody retrieval with microwave for 3,5, or 10 minutes. Is it true tissue were treated for 23 minutes. I am afraid this is too high it might damaged the tissue which is why antibody is binding non specifically.

      - You have mentioned that "staining is OK" with Lymph nodes, this indicate the quality of ab is fine so the problem might be with protocol. Please troubleshoot the protocol as required.

      The following publication show nice IL-17 staining. Could you send us your image?
