Biological function summary
ICP8 interacts with other viral and cellular proteins to organize the replication compartment inside the host cell nucleus. It forms part of the replication complex working alongside HSV-2 DNA polymerase and other accessory proteins to effectively duplicate the viral genome. ICP8 facilitates the unwinding of DNA and stabilizes the single-stranded conformation ensuring that the replication is efficient. Its ability to bind single-stranded DNA and engage with various partners highlights its importance in managing viral replication dynamics.
Pathways
ICP8 is central to the HSV-2 replication and DNA synthesis pathways. The protein acts in concert with viral proteins such as UL42 and HSV DNA polymerase to enhance replication efficiency. The DNA repair pathway also sees involvement by ICP8 allowing interplay with cellular mechanisms to support or hinder the cellular response to viral assault. ICP8 ensures that HSV-2 effectively commandeers the host's cellular machinery to prioritize viral genome amplification.
