Herpes Simplex Virus 1 (HSV1) also referred to as HSV-1 is an enveloped virus with a large double-stranded DNA genome of about 152 kb. HSV1 possesses a complex architecture featuring numerous glycoproteins including gHSV1 that play significant roles in its entry and immune evasion. This virus predominantly targets epithelial cells in the oral genital regions and sensory neurons. HSV1 expresses various proteins vital for its replication and viral particle assembly within host cells.
Biological function summary

HSV1 interacts with the host's cellular mechanisms to establish latency and persistent infection. During infection several HSV1 proteins contribute to its ability to subvert and evade the host immune response sometimes forming part of existing viral protein complexes. Virion-host cell membrane fusion facilitated by glycoprotein gHSV1 is an important step allowing the viral genome to enter the host cell driving subsequent viral replication.

Pathways

The life cycle of HSV1 includes entry replication and egress tightly integrated into cellular pathways like the apoptosis and the interferon response pathways. HSV1 modulates the apoptotic pathway through proteins such as ICP22 which allows survival and efficient viral replication. Also it influences the interferon response pathway by inhibiting the host's antiviral mechanisms ensuring viral persistence in the host.

HSV1 is most commonly known to cause oral herpes and can lead to encephalitis in severe cases. The virus reactivates from latency within neurons potentially leading to periodic disease recurrence. During infection HSV1 interacts with host proteins such as TNF-alpha which can contribute to inflammatory responses and ICP0 which facilitates viral replication and disrupts host cell function. Understanding HSV1's mechanisms and pathways is critical for developing effective anti-HSV-1 antibodies and therapeutic strategies.
