HMW Cytokeratin also known as High Molecular Weight Cytokeratin refers to a group of keratin proteins typically around 66-68 kDa that forms part of the cytoskeleton in epithelial cells. These proteins express mainly in stratified squamous epithelia and some simple epithelia providing structural integrity and resistance to mechanical stress. They occur in epithelial tissues like the skin mouth esophagus and bladder playing a role in cellular differentiation and tissue organization.
Biological function summary

High molecular weight cytokeratins contribute significantly to the structural framework of cells. They form intermediate filaments that provide mechanical support and maintain cell shape. As part of this filament complex HMW Cytokeratins interacts with other cytoskeletal structures and adhesion molecules taking an active part in cellular dynamics and stability. These keratins also assist in the protection of epithelial cells from mechanical and non-mechanical stress.

Pathways

HMW Cytokeratins plays an important role in the keratinization pathway and epithelial cell differentiation processes. In the keratinization pathway these proteins interact with other keratins like KRT5 and KRT14 helping in the formation of resilient and tough surfaces on epithelial cells. In epithelial cell differentiation they are important in the regulation of cellular processes that define cell fate and tissue structure engaging with proteins like p63 which regulates gene expression in epithelial stem cells.

High molecular weight cytokeratins associates with several epithelial-derived cancers including squamous cell carcinoma and skin-related disorders such as ichthyosis. Aberrant expression or mutation in these keratin genes can lead to impaired epithelial cell function or transformation. In certain cancers HMW Cytokeratins often coexpress with proteins like vimentin indicating epithelial-mesenchymal transition and aiding in cancer diagnosis and prognosis.

Regulates the recruitment of Langerhans cells to the epidermis, potentially by modulation of the abundance of macrophage chemotactic cytokines, macrophage inflammatory cytokines and CTNND1 localization in keratinocytes (By similarity).
