HIC2 also known as Hypermethylated in Cancer 2 acts mechanically as a transcriptional repressor. It belongs to the bHLH-PAS family of transcription factors and exhibits a molecular mass of approximately 52 kDa. The protein is ubiquitously expressed with significant expression in heart and skeletal muscle tissues. HIC2 contains a bHLH domain responsible for DNA binding and dimerization facilitating its role in transcriptional repression.
Biological function summary

HIC2 influences gene expression by interacting with other transcription factors. It binds to specific DNA sequences regulating genes involved in cellular differentiation and development. HIC2 often functions within a larger protein complex integrating signals that affect cellular response and proliferation. Due to its regulatory role it plays a part in managing cell cycle and maintaining cellular homeostasis.

Pathways

HIC2 integrates into critical signaling pathways that govern cell growth and differentiation. It participates in the Notch signaling pathway contributing to the regulation of cell fate decisions. HIC2 is also associated with the Hippo pathway which controls organ size and tissue regeneration. These pathways involve interaction with related proteins such as YAP1 in the Hippo pathway and RBPJ in the Notch pathway.

HIC2 displays significant relevance to cancer and congenital heart defects. Altered HIC2 expression or function may contribute to tumorigenesis as it plays a role in cell cycle regulation and apoptosis. Additionally mutations or dysregulation in the HIC2 gene have links to congenital heart defects. Its connections to proteins like TAZ in cancer progression further highlight the importance of HIC2 in disease contexts.
