Biological function summary
NS3 plays a significant role in the life cycle of Hepatitis C Virus by facilitating viral replication and maturation. NS3 protease activity cleaves the non-structural protein precursors at multiple sites which is essential for the formation of the viral replication complex. As part of the HCV replication complex it associates with the NS4A cofactor that enhances its protease function. The protein is highly conserved and represents an attractive target for antiviral drug development due to its indispensable role in the viral life cycle.
Pathways
NS3 is integral to the viral replication and host immune evasion pathways. The protein interacts in the post-translational modification processing pathway that is critical for producing functional viral proteins. NS3 activity is closely related to other non-structural proteins such as NS5A and NS5B which are involved in RNA replication. These interactions form a network essential for the hepatitis C infection propagation influencing the virus's ability to hijack host cells' functions.
