    The spiral shaped bacterium Helicobacter pylori is strongly associated with inflammation of the stomach and is also implicated in the development of gastric malignancy. H. pylori is known to cause peptic ulcers and chronic gastritis in human. It is associated with duodenal ulcers and may be involved in development of adenocarcimona and low-grade lymphoma of mucosa associated lymphoid tissue in the stomach. More recently this bacterium has also been implicated with a number of vascular disorders including heart disease. It is not clear how H. pylori is transmitted or why some patients become symptomatic while others do not. The bacteria are most likely spread from person to person through fecal-to-oral or oral-to-oral routes. Possible environmental reservoirs include contaminated water sources. Serological tests that measure specific H. pylori IgG antibodies can determine if a person has been infected although these methods do have drawbacks and limitations.

DESCRIPTION OF THE PROBLEM No signal. I used a dilution of 1/120000 as mentioned in the technical sheet, but I get no signal. Is it really that dilution 1/120000?

TYPE OF ELISA cells were coated on 96 cell culture plates (used for ELISA). Bacteria were incubated with the cells. After wash step and blocking step, primary antibody (rabbit polyclonal, abcam 20459 (1/5000) against bacteria was added, then secundary (ab6721, 1/120000), then ABTS was added. result: no signal. What did I do wrong?

Reviewing this case, I would like to offer some suggestions to help optimise the results fromthese antibodies.I would also appreciate if you can confirm some further details:

1. Please confirm the order number and date of purchase? I am sorry the last order from Belgium for this antibody was over 1 year ago.Also, Iam sorry to confirm that ab20459 Anti-Helicobacter pylori antibody jas not been tested in this type of cell based ELISA, it has been tested in indirect ELISA only.Regrettably, this would no longer be covered by the 6 month guarantee, or guaranteed for this application.

2. As the protocol uses cells coated on the plates and then incubated with the H. pylori organisms. The Tween in the buffers may be dissolving the cell membranes. It is also possible that the number of H. pylori organisms per well are too low for detection,manymay be washed away during the procedure.

3. In testing, the Helicobacter pylori isdirectly grownin the microtiter wells. A suspension of the organisms (barely hazy) is dispensed to the wells (50-100uL per well). The plates are air-dried at room temperature overnight or until dry. The plates should then be washed with a buffer containing Tween 20 to remove any loosely bound organisms before running the assay.

There is also a published reference from another researcher using this antibody in a nanoparticle assay. I have included a link to this paper. I hope you may find this helpful:
