  • 機能

    Responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4). Histone deacetylation gives a tag for epigenetic repression and plays an important role in transcriptional regulation, cell cycle progression and developmental events. Histone deacetylases act via the formation of large multiprotein complexes. Deacetylates SP proteins, SP1 and SP3, and regulates their function. Component of the BRG1-RB1-HDAC1 complex, which negatively regulates the CREST-mediated transcription in resting neurons. Upon calcium stimulation, HDAC1 is released from the complex and CREBBP is recruited, which facilitates transcriptional activation. Deacetylates TSHZ3 and regulates its transcriptional repressor activity. Deacetylates 'Lys-310' in RELA and thereby inhibits the transcriptional activity of NF-kappa-B.

    Answer

    Thank you for your enquiry. This antibody is sold as whole antiserum. The IgG concentration has not been determined. However, I would expect it to be within a range of 10-16mgml-1 given that it is rabbit whole serum. We have not received reviews of this product. Please can you detail where you have seen details of the concentration of this antibody? This antibody has been applied by chromatin IP in the following publication; Breiling A et al. General transcription factors bind promoters repressed by Polycomb group proteins. Nature 412:651-5 (2001). PubMed: 11493924 I hope this information helps, please do not hesitate to contact us if you need any more advice or information.

    Question

    I want to do ChIP with the HDAC1 antibody. I would like to know if western results show non specific bands being recognised. Is there a reference for this antibody, has it been used in any published papers? Could you send me some of your test data for western and ChIP with this antibody? Dilutions at which it has worked for you? conditions for western and ChIP? Could you send me a small aliquot to test. I have brought histone antibodies from you before. One of them works great, the other doesnt. Anyway, any information would be of great help.Thanks

    Thank you for your enquiry. All the information that we have regarding ab1767 is located on the online datasheet. Unfortunately, we are not aware of any publications that feature the use of this antibody. To our knowledge, customized protocols are not required for this antibody. For Western blotting, I would suggest starting at a dilution of 1:100. We do not routinely offer free or trial sized samples for testing purposes. Our policy at Abcam is that if an antibody does not work as specified on the datasheet, we will offer a replacement or reimbursement.

    Question

    I ordered the anti-HDAC1 antibody (ab1767) that is supposed to be reactive in Drosophila. I performed western blotting on whole Drosophila heads and, at three different dillutions, and obtained multiple strong bands. I would like to know what is the size that has been determined to be HDAC1 (Rpd3) and what are the conditions used for performing such a western blot. Thank you.
