Flow Cytometry (Intracellular) - Anti-HAPLN1 antibody [EPR6338] - BSA and Azide free (AB240257)
Intracellular Flow Cytometry analysis of MCF-7 (human breast carcinoma) cells labeling HAPLN1 with purified ab181997 at 1/2000 dilution (red). The secondary antibody was Goat anti rabbit IgG (Alexa Fluor® 488) at 1/2000 dilution. A Rabbit monoclonal IgG (Black) was used as the isotype control and cells without incubation with primary antibody and secondary antibody (Blue) were used as unlabeled control.
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab181997).

Immunocytochemistry/ Immunofluorescence - Anti-HAPLN1 antibody [EPR6338] - BSA and Azide free (AB240257)
Immunofluorescent analysis of MCF7 cells (paraformaldehyde-fixed, 4%) labeling HAPLN1 with ab181997 at 1/250 dilution, followed by Goat anti rabbit IgG(Dylight 488) at 1/200 dilution and counter-stained with DAPI (blue).
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab181997).

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-HAPLN1 antibody [EPR6338] - BSA and Azide free (AB240257)
Immunohistochemical analysis of paraffin-embedded Human transitional cell carcinoma of bladder tissue labeling HAPLN1 with ab181997 at 1/50 dilution followed by pre-diluted HRP-conjugated secondary antibody and counter-stained with Hematoxylin.
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab181997).
Perform heat mediated antigen retrieval with EDTA buffer pH 9 before commencing with IHC staining protocol.

製品の詳細
ab240257 is the carrier-free version of ab181997.
Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.
What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:
- - High batch-to-batch consistency and reproducibility
- - Improved sensitivity and specificity
- - Long-term security of supply
- - Animal-free batch production
For more information, read more on recombinant antibodies.
Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.
Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.
Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Biological function summary
Hyaluronan and proteoglycan link protein 1 supports the organization and stabilization of the extracellular matrix network. As part of this framework HAPLN1 forms complexes with hyaluronan and aggrecan ensuring structural stability and resilience in tissues experiencing mechanical stress. The association with such complexes impacts cellular signaling and tissue hydration which are important for maintaining tissue homeostasis and function.
Pathways
HAPLN1 participates in extracellular matrix organization and cell signaling processes. It is actively involved in the aggrecan pathway which influences cartilage development and repair. HAPLN1 is also connected to proteins like aggrecan and link proteins that help mediate interactions within the matrix and regulate cellular responses to mechanical stress.
