The G protein-coupled receptor 52 (GPCR GPR52) also known simply as GPR52 functions as a receptor within the central nervous system. It encodes for a protein that weighs around 42 kDa. This receptor is part of the larger GPCR family which is known for mediating various physiological responses. GPR52 mainly expresses in the brain particularly in regions like the striatum and cortex areas important for cognition and motor control.
Biological function summary

This receptor facilitates neurotransmission by modulating cAMP (cyclic adenosine monophosphate) levels when activated. It does not require a complex formation but acts directly in this signaling process. It affects the release of neurotransmitters influencing activities such as movement motivation and emotion. Dopaminergic signaling which ties to GPR52 activity plays a critical role in these functions.

Pathways

GPR52 integrates into the dopamine signaling and cAMP signaling pathways. It couples with Gs protein leading to increased intracellular cAMP levels upon activation. Its relationship with the dopamine D2 receptor is significant as GPR52 modulates the dopaminergic system therefore influencing various neural pathways linked to mood and behavior regulation.

Gs-coupled receptor activated by antipsychotics reserpine leading to an increase in intracellular cAMP and its internalization (PubMed : 24587241). May play a role in locomotor activity through modulation of dopamine, NMDA and ADORA2A-induced locomotor activity. These behavioral changes are accompanied by modulation of the dopamine receptor signaling pathway in striatum (PubMed : 24587241). Modulates HTT level via cAMP-dependent but PKA independent mechanisms throught activation of RAB39B that translocates HTT to the endoplasmic reticulum, thus avoiding proteasome degradation (PubMed : 25738228).
