補足情報

This supplementary information is collated from multiple sources and compiled automatically.

GORASP2 also known as GRASP55 is a golgi reassembly stacking protein with a mass approximately 55 kDa. It is highly expressed in the Golgi apparatus where it plays an important role in maintaining the structure and function of this organelle. GRASP55 facilitates the stacking of Golgi cisternae which is essential for Golgi apparatus organization. The protein acts alongside its isoform GRASP65 though they function somewhat independently in different Golgi regions.
Biological function summary

GRASP55 controls various cellular processes including the trafficking of proteins and lipids within the cell. It is part of the GRASP complex along with GRASP65 facilitating membrane tethering and fusion events. The protein's function is also linked to its ability to regulate Golgi's response to cellular stress or changes in environment impacting secretion and growth factor availability. In some conditions GRASP55 can influence processes such as autophagy underlining its diverse biological actions within the cell.

Pathways

GRASP55 integrates into significant cellular pathways like the secretory pathway and the autophagy pathway. It interacts with key proteins like golgins and Rab GTPases to assist in vesicle tethering and transport. GRASP55 also closely interacts with p62 a protein that plays a critical role in the selective autophagy pathway. This positions GRASP55 as a contributor to not only the routine processing of cellular materials but also cellular adaptation to stress through autophagy modulation.

GRASP55 shows relevance to cancer progression and neurodegenerative diseases. Aberrant expression or dysfunction of GRASP55 can contribute to cancerous transformations by altering cell adhesion and signaling pathways. Its role in autophagy connects it to diseases like Alzheimer's where defective protein processing is a hallmark. The interaction of GRASP55 with proteins such as p62 becomes particularly important as disrupted autophagy pathways are often implicated in neurodegenerative conditions highlighting GRASP55's significant role in both health and disease.

Key structural protein of the Golgi apparatus (PubMed : 33301566). The membrane cisternae of the Golgi apparatus adhere to each other to form stacks, which are aligned side by side to form the Golgi ribbon (PubMed : 33301566). Acting in concert with GORASP1/GRASP65, is required for the formation and maintenance of the Golgi ribbon, and may be dispensable for the formation of stacks (PubMed : 33301566). However, other studies suggest that GORASP2 plays a role in the assembly and membrane stacking of the Golgi cisternae, and in the process by which Golgi stacks reform after breakdown during mitosis and meiosis (PubMed : 10487747, PubMed : 21515684, PubMed : 22523075). May regulate the intracellular transport and presentation of a defined set of transmembrane proteins, such as transmembrane TGFA (PubMed : 11101516). Required for normal acrosome formation during spermiogenesis and normal male fertility, probably by promoting colocalization of JAM2 and JAM3 at contact sites between germ cells and Sertoli cells (By similarity). Mediates ER stress-induced unconventional (ER/Golgi-independent) trafficking of core-glycosylated CFTR to cell membrane (PubMed : 21884936, PubMed : 27062250, PubMed : 28067262).
