    Question

    Actually, ab53942 is the only one thatI have known reacting with rabbit, I have tried some other antioxidantenzymes such as HO-1andNQO1 but they don't have reactivity with rabbit so they didn't work.We don't want to try any other new antibody in case it is not certain to react with rabbit, our material is very precious since this is an animal experiment and also our time is precious too. If you are able to homology check of your antibodies such as HO-1, NQO1, GST, GCLM, GCLCwith rabbit,may beuseful for our study. And please let me know about the exact time for replacement for ab53942 when you know about it. Thank you for your attention.

    I have found two antibodies for rabbit samples that may be effective for detecting two of the proteins you specified. Anti-GCLC ab80841 is raised against an immunogen sequence that is conserved 100% in the rabbit sequence listed in the UniProt entry for accession G1T442: http://www.uniprot.org/uniprot/G1T442 The immunogen sequence for ab80841 is CRWGVISASVDDRTREERG. The datasheet is at the following URL: Click here (or use the following: https://www.abcam.com/GCLC-antibody-ab80841.html). For HO-1, I found ab82585: Click here (or use the following: https://www.abcam.com/Heme-Oxygenase-1-antibody-ab82585.html). I have requested the rabbit data from the originating laboratory and will forward this when I receive it.. Both of these are rabbit polyclonals, so a secondary anti-rabbit IgG antibody will likely stain denatured and reduced endogenous rabbit IgG at 25kDa and 50kDa for the light and heavy IgG subunits, but you should still be able to clearly distinguish the signal from the protein of interest. I am looking into obtaining a different lot of ab53942 and hope to have an update for you tomorrow. In the meantime, if you have any questions, please do not histiate to ask.

    Question

    I prepare my samples as follows: Firstly, our groups are i)control, ii)cholesterol diet,iii) cholesterol+vitamin E treatment. My blotting results are in 3 sets. We homogenize our rabbit aortic tissues with homojenization buffer. Then the samples mixed with sample buffer including mercaptoethanol and bromphenolblue. The samples then denatured at 95 C for 5 minutes. Denatured samples are loaded to %10 stacking and %5 running gel. This is the protocol I have been doing for two years. I couldn't exactly understand what do you mean if they are reduced. Dont hesisate to ask further questions if my answer is not enough.

    Thank you for your sample preparation protocol. It is standard and much as I expected. What I meant by "reduced" was whether you had included a reducing agent in the loading buffer, to reduce di-sulfide bonds. The GST apparently exists as a di-sulfide-linked dimer, in the soluble cytosolic form, and I assume that is why some people observe it at 45-50 kDa. However, you do include mercaptoethanol in your sample buffer, and I am assuming it is sufficient to reduce any di-sulfide bonds. We expect the band for GST monomer to appear at about 25-27 kDa. I do not have an explanation for the bands at 35 kDa, and for that matter, I do not understand why the standard in the blot shown on the datasheet runs at about 33 kDa. (I have asked the lab; it may have a tag). For an example of a blot that shows a signal at the expected molecular weight, please see the datasheet for ab27489: Click here (or use the following: https://www.abcam.com/GSTM1--GSTM2-antibody-ab27489.html). (The immunogen unfortunately does not have any homology with rabbit GST mu1 described at the UniProt entry http://ebi1.uniprot.org/uniprot/P46409 ).     Question

    I am working with Abcam GST antibody with a product number:53942. I am working with rabbit aorta and using Western Blotting technique. My question is: in antibody's datasheet, Abcam reported that the bands should be observed at 33 kDa. But in my blots, I have been observing two bands which are at 35 kDa and 50 kDa (which is even more notable than 35 kDa sized one). But you haven't implied any nonspecific bands in the datasheet. As you know Nrf2 regulates GST expression. And also GST cytosolic forms are between 45-55 kDa. So I wonder what is that 50 kDa bands in my samples?
