機能
Ionotropic glutamate receptor. L-glutamate acts as an excitatory neurotransmitter at many synapses in the central nervous system. Binding of the excitatory neurotransmitter L-glutamate induces a conformation change, leading to the opening of the cation channel, and thereby converts the chemical signal to an electrical impulse. The receptor then desensitizes rapidly and enters a transient inactive state, characterized by the presence of bound agonist. -

Purified ab32436 at 1/50 dilution (2µg) immunoprecipitating Glutamate Receptor 1 (AMPA subtype) in SH-SY5Y whole cell lysate.
Lane 1 (input): SY-SY5Y (Human neuroblastoma epithelial cell) whole cell lysate 10µg
Lane 2 (+): ab32436 + SH-SY5Y whole cell lysate.
Lane 3 (-): Rabbit monoclonal IgG (ab172730) instead of ab32436 in SH-SY5Y whole cell lysate.
VeriBlot for IP Detection Reagent (HRP) (ab131366) (1/1000 dilution) was used for Western blotting.
Blocking Buffer and concentration: 5% NFDM/TBST.
Diluting buffer and concentration: 5% NFDM/TBST.
Observed band size: 106 kDa -

Immunohistochemical analysis of Human hippocampus tissue, staining Glutamate Receptor 1 (AMPA subtype) with ab32436.
Antigen retrieval was performed by heat mediation in a citrate buffer (pH 6). Samples were blocked with 10% normal goat serum for 30 minutes before incubating with primary antibody (1/20) for 2 hours. A biotinylated anti-rabbit IgG was used as the secondary antibody and staining was detected using DAB.
