      Overlay histogram showing HepG2 cells stained with ab85715 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab85715, 0.5µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in HepG2 cells fixed with 80% methanol (5 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.

    Question

    Thank you for providing me the information on the pancreas tissue lysate ab29816 (lot # GR26918-2). Based on our conversation by phone in order to try to determine the cause of the problem, I agreed to run the pancreas tissue lysate in a gel and stained it with Coomassie blue before and after the transfer. The results are attached. Since I did have very limited materialleft, I was only able to add 10 ug of lysate per lane. I also added the liver tissue lysate (Abcam ab29889 lot GR67555-1) that was managed under the same conditions than the pancreas tissue.

    After the staining with Coomassie blue, the results showed that I can clearly see the material in the liver sample, but I was unable to see it in the pancreas. The amount of material in the pancreas sample is clearly significantly less than the liver sample and than the 3.75 ug/uL that is on the label. Something that I corroborated with other samples prepared in house. This clearly affecting to determination of the reactivity of the antibody anti-GluT2 (ab95256) antibody, where 10 ug are recommended to detect the protein by Western blot.

    We have purchased twice this material and it has failed each time. Can we get a replacement of other pancreas lysate lot? If that is not the case what can we do?

    Thank you for your reply and also for checking the lysate. This is really helpful.

    I am sorry to hear that the problem has not been resolved.
    Since you see no protein bands before transfer, it does not surprise me that you see none after. I, too, think that the lysate should give you bands at 10 ug.

    I have checked what we have in stock for ab29816: It is only the same lot as you have tried.
    Also, this is the only human normal pancreas tissue whole cell lysate. All other human pancreas lysates are either disease-related or organelle specific (membrane, nuclear).

    If the antibody ab95256 works well with the liver lysate, I'd suggest to use this lysate as positive control.

    I'd be happy to credit or refund the purchases of the 2 vials of pancreas lysate ab29816 - unless you would like to replace them with another, differentlysate that we carry. Please let me know what would be best for you.

    I am sorry to hear about the problem the pancreas lysate is causing you.

    As we discussed over the phone, it would be worthwhile trying the following in order to determine the cause of the problem:

    1)stain gel before and after transfer with Coomassie blue (protein should be detected)
    2)use more protein (e.g. 10 ug, 20 ug or 50 ug) - if feasible

    I have also contacted the lab tocheckfor more information about this lotand additional suggestions. I will be in touch as soon as I hear back from them.

    Vielen Dank für Ihre Anfrage.

    Leider haben wir keinen Antikörper, der allen genannten Anforderungen entspricht.

    Wir führen einen Maus-monoklonalen GLUT2-Antikörper, der bislang nur für humane Proben getestet wurde (ab85715). Das Immunogen zeigt eine Sequenzähnlichkeit von 81% gegenüber dem murinen GLUT2, und daher könnte der Antikörper mit Maus kreuzreagieren.

    Daneben haben wir zwei GLUT2-Antikörper, die wir für die Anwendung in der Maus garantieren können (ab54460, ab104622). Jedoch sind die beiden polyklonale Antikörper aus dem Kaninchen.

    Leider ist auch keiner unserer GLUT2-Antikörper mit Biotin markiert. In der Regel würde ich daher einen unsere EasyLink-Konjugationskits empfehlen (ab102863 bzw. ab102866, https://www.abcam.com/EasyLink). Da Sie jedoch beträchtliche Mengen des Antikörpers benötigen, ist das sicher nicht praktikabel. Leider können wir derzeit auch keinen Konjugationsservice auf Anfrage für grössere Mengen anbieten.

    Zudem habe ich die Vermutung, dass Sie den Antikörper vielleicht für funktionelle Studien verwenden möchten, weil Sie nach 18 mg fragten. Leider ist unseres Wissens bislang keiner der GLUT2- Antikörper für so etwas getestet worden, und wir können daher nicht garantieren, dass es mit dem jeweiligen Produkt funktioniert. Wir geben alle getesteten Anwendungen auf unseren Produktdatenblättern an und aktualisieren diese sobald wir neue Informationen erhalten. Falls sich herausstellt, dass der Antikörper nicht so funktioniert, wie auf dem Datenblatt beschrieben und Sie sich innerhalb von sechs Monaten mit Ihrem Problem an uns wenden, werden wir Ihnen gerne Vorschläge zur Optimierung Ihres Protokolls oder gegebenenfalls einen Ersatz oder eine Gutschrift schicken.
