Immunocytochemistry/ Immunofluorescence - Anti-GHRHR antibody (AB76263)

Immunofluorescence analysis of mouse pituitary cells, staining GHRHR with ab76263.

Cells were fixed with 4% paraformaldehyde and blocked using 4% NGS in PBS with 0.3% Triton X-100 for 1 hour at room temperature. Cells were incubated with primary antibody (1/50) in 4% NGS for 18 hours at 4°C. Slides were washed three times for 15 min with PBS and then incubated in 4% NGS with an AlexaFluor®488-conjugated anti- rabbit secondary antibody (1/1000) for 1 hour at room temperature.

補足情報

This supplementary information is collated from multiple sources and compiled automatically.

GHRHR also known as the growth hormone-releasing hormone receptor is an important protein that plays an important role mechanistically in the human body. This receptor is located on the cell membrane and binds to growth hormone-releasing hormone (GHRH). GHRHR is expressed predominantly in the pituitary gland. It has a molecular mass of approximately 44 kDa and its activation leads to the intracellular signaling cascades that promote growth hormone secretion.
Biological function summary

The interaction of GHRHR with GHRH is necessary for the regulation of growth hormone release. This receptor is not part of a larger complex; instead it functions individually as a receptor. GHRHR's role in stimulating growth hormone release makes it essential for normal growth metabolism and body composition. It influences processes like protein synthesis lipolysis and the maintenance of normal glucose levels.

Pathways

GHRHR operates within the growth hormone signaling pathway. This pathway involves the activation of GHRHR by GHRH which leads to the stimulation of adenylate cyclase increasing cyclic AMP levels resulting in growth hormone release. This pathway is interconnected with other signaling proteins like somatostatin which acts as an inhibitor of growth hormone secretion providing a balance in growth regulation.

GHRHR has been linked to conditions affecting growth such as dwarfism and gigantism. Mutations in the GHRHR gene can disrupt normal growth hormone signaling leading to reduced or excessive growth. Gigantism involves increased growth hormone secretion often related to pituitary gland abnormalities. In contrast dwarfism may result from insufficient growth hormone action due to receptor defects. GHRHR's interaction with GHRH and its role in growth hormone regulation highlight its significance in these growth-related disorders.
