Anti-Ghrelin Receptor/GHS-R 抗体 (ab85104)

Antibody ab92520 is raised against a ghrelin receptor amino acid sequence that is found in both isoforms of the human receptor, so it should be capable of recognizing both. Antibody ab85104 is raised against a sequence derived from rat receptor that is 80% conserved in both human isoforms. It will probably recognize the human isoforms but the probability is lower than for ab92520.

The isoforms 1A and 1B differ in amino acid 266-289 and 290-366, as these are missing in Isoform 1B. http://www.uniprot.org/uniprot/Q92847. Based on this data antibody ab125457 would detect the Isoform 1A only.

ab85104, ab14480, ab14481, ab95250 etc would detect both the isoforms.

There is requirement for secondary antibodies if you are planning to use these un-conjugated antibodies in WB, ELISA, ICC/IF or immunohistochemistry. You might not need secondary antibody with ab66406, ab48285.

Unfortunately, we haven't tested if this ab85104 Anti-Ghrelin Receptor antibody can distinguish between the GHS-R1a and GHS-R1b protein. However, as the immunogen is derived from the conserved N-terminus of both forms, it is possible that the antibody cannot distinguish those receptors, and therefore, detects both forms.
Regarding the ab9749 Anti-Leptin antibody, I am wondering if you have purchased this product over one of our official distributors Biogen or Sellex? Do you remember if you received 200ul? Indeed the price seems very high, and I am surprised that you were only able to use it on 6 slides out of a 200ul product (seen that you dilute this antibody approximately at 1/250).
In addition, it looks like that this Leptin antibody has only been tested on mouse so far, but not on human samples. This, unfortunately, means that we cannot guarantee that the antibody will be working on human samples in the first place. We can see whether we could provide you with a testing discount as we do not carry any test samples of our antibodies.
However, we need the additional information we are requesting from you and you would need to contact then our official distributor.
The ab85104 Anti-Ghrelin Receptor antibody is already tested on both application and species and therefore not appropriate for this offer.

I just wanted to confirm that you were testing reactivity of the antibody with protein based on the rat or human sequence. As you confirmed you are working with human protein it does appear there is an issue with the vial of ab85104 in this case.

I just want to confirm that your cDNA for GHSR-1 is based on human or rat sequence, as those are the tested species to date.

Just a few additional questions:
1) Is a full length GHSR-1 being expressed or is the consturct a fragment of GHSR-1?
2) What species of cDNA was used to produce the GHSR-1 clone?
3) Are you able to determine recombinant protein has been expressed by any other means (eg. fusion protein or epitope tag)
4) Would it be useful to run the gel under reducing conditioins?
