Gbx2 also known as Gastrulation Brain Homeobox 2 functions as a transcription factor. This protein approximately 24 kDa in mass plays a significant role in gene regulation during embryonic development. Gbx2 is expressed mainly in the developing nervous system particularly in the hindbrain and midbrain regions. It influences the expression of other genes critical for brain and cranial nerve development.
Biological function summary

The role of this transcription factor centers on regulating neural development processes. Gbx2 participates in formation and patterning of the central nervous system. It does not typically operate alone; instead it often interacts with other transcription factors to modulate gene expression. Gbx2 is important for demarcating boundaries in the hindbrain which contributes to proper compartmentalization and formation of rhombomeres.

Pathways

Gbx2 contributes significantly to the Wnt and FGF signaling pathways which are fundamental during early neural development stages. In these pathways Gbx2 interacts closely with other key proteins such as Otx2 another homeobox gene to establish clear anterior-posterior axis in the developing brain. These interactions are essential to ensure proper differentiation and positioning of neural cells.

Such abnormalities may associate with disorders like Joubert Syndrome where proper brain segmentation does not occur. Additionally Gbx2's role connects it to the Shh pathway where imbalances might exacerbate conditions like medulloblastoma a type of brain cancer found in children through interactions with proteins such as OTX2.
