Immunocytochemistry/ Immunofluorescence - Anti-gamma Catenin antibody [15/γ-Catenin] - BSA and Azide Free (AB305242)
This data was developed using ab305241, the same antibody clone in a different buffer formulation. Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized MCF7 (human breast adenocarcinoma epithelial cell) cells labeling gamma Catenin with ab305241 at 1/100 dilution (9.85 µg/ml), followed by ab150117 Goat Anti-Mouse IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 dilution (2µg/ml) (Green). Confocal image showing cell-junction staining in MCF7 cell line. Low expression : THP-1 (PMID : 22858986). Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8). ab179513 Anti-beta Tubulin rabbit monoclonal antibody was used to counterstain tubulin at 1/200 dilution (10µg/ml) followed by ab150080 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 594) at a 1/500 dilution (4µg/ml) (Red). The nuclear counterstain was DAPI (Blue). -ve control 1 : ab305241 at 1/100 dilution followed by at ab150080 a 1/500 dilution. -ve control 2 : ab179513 at 1/200 dilution followed by at ab150117 a 1/1000 dilution.

This data was developed using ab305241, the same antibody clone in a different buffer formulation.
gamma Catenin was immunoprecipitated from 0.35 mg MCF7 (human breast adenocarcinoma epithelial cell) whole cell lysate 10 µg with ab305241 at 1/30 dilution (2µg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab305241 at dilution. VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution.
Blocking and dilution buffer and concentration : 5% NFDM/TBST.

Biological function summary
Gamma Catenin is an integral component of the armadillo family of proteins and part of desmosomal and adherens junction complexes. It plays an essential role in maintaining the integrity and mechanical strength of tissues. Through interacting with other proteins in these complexes gamma Catenin facilitates cell signaling and communication. It associates with cadherins and catenins including plakophilin and β-catenin to ensure proper cell-cell adhesion and signaling across the cell membrane.
Pathways
Gamma Catenin is involved in the Wnt signaling pathway an important route governing cellular processes such as proliferation differentiation and migration. Gamma Catenin interacts with β-catenin in this pathway influencing transcriptional regulation in the nucleus. It also plays roles in the cadherin-mediated cell adhesion pathway working alongside other proteins like E-cadherin to modulate cell behavior. These pathways highlight its importance in cellular architecture and signaling.
