Fyn and Yes1 are Src family kinases key players in signal transduction processes within cells. They share similarities in function and mechanism often grouped together due to their roles in cellular phosphorylation events. Fyn and Yes1 have a molecular mass of approximately 59 kDa and are ubiquitously expressed in many tissues including brain thymus and T lymphocytes. Fyn known also as p59fyn contributes significantly to the regulation of immune response and neuronal signaling by phosphorylation of different substrates.
Biological function summary

Fyn and Yes1 influence various cellular processes such as cell growth differentiation and survival. They form part of the larger signaling complexes that regulate cytoskeleton organization helping in maintaining cell structure and motility. When activated they communicate changes in the cellular environment by phosphorylating tyrosine residues on target proteins affecting downstream signaling pathways.

Pathways

Both Fyn and Yes1 play major roles in the mitogen-activated protein kinase (MAPK) pathway and the phosphoinositide 3-kinase (PI3K) pathway. These pathways are fundamental for transmitting extracellular signals to the cell nucleus triggering responses like cell proliferation or survival. Through these pathways Fyn and Yes1 interact with various proteins including phospholipase C-gamma (PLCγ) and SH2 domain-containing proteins. Their activation leads to further signaling cascades that impact cellular outcomes.

Aberrant Fyn and Yes1 activity have been linked to cancer and neurodegenerative disorders. Overexpression or mutation of these kinases can lead to uncontrolled cell proliferation contributing to breast cancer. In the context of neurodegenerative diseases Fyn interacts with the amyloid precursor protein implicated in Alzheimer's disease. Inactive FYN is phosphorylated on its C-terminal tail within the catalytic domain (PubMed : 15489916). Following activation by PKA, the protein subsequently associates with PTK2/FAK1, allowing PTK2/FAK1 phosphorylation, activation and targeting to focal adhesions (PubMed : 15489916). Involved in the regulation of cell adhesion and motility through phosphorylation of CTNNB1 (beta-catenin) and CTNND1 (delta-catenin) (PubMed : 17194753). Regulates cytoskeletal remodeling by phosphorylating several proteins including the actin regulator WAS and the microtubule-associated proteins MAP2 and MAPT (PubMed : 14707117, PubMed : 15536091).
