I would like to offer additional suggestions based on this new information which may help you achieve good results using ab105915. When performing western blot on mitochondrial proteins there are a number of steps which our Mitosciences team suggest to create the best results.

For performing Western blotting transfer and detection, it is always recommended toisolate mitochondria from cells before analysis. Protocols for mitochondrial isolation can be found athttps://www.abcam.com/index.html?pageconfig=resource&rid=11411. While it is possible to probe whole tissueor cell extract, this may result in a weaker signal and/or additional bands resulting from non-specific cross-reactivity to non-mitochondrial proteins. Such as you may be seeing. Additionally we do offer mitochondria isolation kits for tissue (ab110169;https://www.abcam.com/Mitochondria-Isolation-Kit-for-Tissue-with-Dounce-Homogenizer-ab110169.htmland ab110168;https://www.abcam.com/Mitochondria-Isolation-Kit-for-Tissue-ab110168.html).



Samples should be solubilized in the SDS-PAGE loading buffer.After solubilization the sample should be spun at 13,000 rpm for 5 minutes. The supernatant should be collected and loaded onto the gel. Note: In most cases thesample need not be heated/boiled in this sample buffer prior to loading. Howeversample heating (95C for 5 min) may be incorporated into this protocol when non specific bands are present. Adding a reducing agent to the sample e.g. DTT, may also help.
