Follicle-stimulating hormone (FSH) is a glycoprotein with a molecular mass of approximately 35 kDa. It is synthesized and secreted by the anterior pituitary gland. FSH sometimes referred to as Follitropin consists of two subunits: the alpha subunit which is common among other pituitary glycoprotein hormones like luteinizing hormone (LH) and thyroid-stimulating hormone (TSH) and the beta subunit which is unique to FSH and imparts its biological specificity. Expression mainly occurs in the pituitary gland and influences various reproductive tissues.
Biological function summary

FSH plays a significant role in the reproductive processes of both males and females. In females it stimulates the growth of ovarian follicles and promotes estrogen production in the ovaries. In males FSH acts on the Sertoli cells of the testes to aid in spermatogenesis. This hormone functions as part of a larger protein complex that interacts with specific FSH receptors on target cells triggering a cascade of intracellular events. These actions involve cyclic adenosine monophosphate (cAMP) as a second messenger and activate related signaling pathways.

Pathways

FSH integrates into the hypothalamic-pituitary-gonadal (HPG) axis a critical regulatory system for reproductive health. FSH is released in response to gonadotropin-releasing hormone (GnRH) and functions in accordance with other related proteins like LH impacting ovarian and testicular function. FSH also participates in the estrogen regulation pathway by modulating estrogen-levels through its action on the ovaries. Interactions with proteins such as inhibin and activin provide feedback control within these pathways.

FSH levels and activity have been linked to conditions such as Polycycstic Ovary Syndrome (PCOS) and hypogonadism. PCOS often involves an imbalance in FSH and LH ratios leading to disrupted follicular development and ovulation issues. In hypogonadism insufficient FSH affects sexual development and reproductive function. FSH-related dysfunctions can also interconnect with other hormonal conditions involving LH and prolactin illustrating its importance in maintaining reproductive health. Researchers use assays like FSH ELISA to measure and study the hormone's levels in these contexts.
